多くのがんで発現が抑制されているがん抑制遺伝子RECKは、マトリックスメタロプロテアーゼ(MMPs)抑制、NotchリガンドのADAM10による放出の制御、Wnt7シグナルの増強などの働きを持つことが既に知られている。我々は、新たに、RECKがマウス胎児脳血管形成においてWnt7シグナルのレセプター側(GPR124-FZD-LRP)だけでなくリガンド側(Wnt7)においてもシグナル伝達にとって必須の役割を担う事を見出した。成体マウスを用いた研究では、RECKが下垂体の成長ホルモン(GH)産生細胞で発現されており、成長ホルモン放出ホルモン受容体(GHRHR)やグレリン受容体(GHSR)の安定化に寄与していること、また、肝臓においても成長ホルモン受容体(GHR)の安定化に寄与している事を発見した。また、RECKはマルファン症候群で変異が見られるフィブリリン1(FBN1)と生体内で共発現しており、膜結合型マトリックスメタロプロテアーゼ(MT1-MMP)、メタロプロテアーゼ(ADAMTS10)と共に、FBN1線維形成に重要な役割を演ずる事を見出した。さらに、皮膚線維芽細胞ではRECKがproMT1-MMPを活性化するのに対し、精製したRECKは活性化型MT1-MMPの活性を抑制するという興味深い現象を見出した。これらの知見は、RECKの生理機能やフィブリリン細毛線維形成の分子機構に新たな光を当て、がん、成長障害、結合組織異常などの病態解明に資するものと考えている。

The tumor suppressor protein RECK has been implicated in the regulation of matrix metalloproteinases (MMPs), Notch-signaling, and WNT7-signaling. We recently found that RECK plays roles essential for vascular development in mouse central nervous system (CNS) not only on the receptor (GPR124-FZD-LRP) side but also on the ligand (Wnt7a/b) side of WNT7-signaling. In adult mouse, RECK is expressed abundantly in growth hormone (GH)-producing cells in anterior pituitary gland. We found that RECK contributes to the stabilization of several receptors involved in the GH-IGF1 axis (i.e., GHRHR, GHSR, GHR). We also found that RECK and fibrillin-1 are coexpressed in many human tissues. Fibrillin-1 mutations are found in Marfan syndrome. Our data suggest that three proteins, RECK, ADAMTS10 (secreted metalloprotease), and MT1-MMP (secreted membrane-bound matrix-metalloprotease) cooperate to support the formation of robust fibrillin-1 fibers. Interestingly, RECK is capable of activating proMT1-MMP in mouse dermal fibroblasts while it inhibits mature MT1-MMP in vitro. These findings shed some new lights on the physiological functions of RECK, the molecular mechanisms of fibrillin fiber formation, and the molecular bases of diseases such as cancer, growth defects, and connective tissue disorders.
