Epigenetic mechanisms of stress-induced depression
研究概要

気分障害の発症には、生物学的要因や心理学的要因に加えてストレスなどの環境要因が関わると指摘されている。すなわち、うつ病患者はストレスに対して脆弱な生物学的素因を有しており、慢性的な環境ストレス暴露により神経可塑性異常を引き起こすことで発症に至る、といったストレス脆弱性仮説である。現在までに、脳画像・遺伝学・分子生物学的手法等を用いて、精神疾患発病脆弱性や危険因子に関わる遺伝・神経生物学的基盤の解析研究が精力的に行われている。一方、慢性ストレスに暴露された人すべてが精神疾患を発症するわけではなく、むしろ大多数の人はストレスに適応して精神的安定性を維持することができる。このようなストレスの影響に対する予防・緩衝要因としてのレジリエンスは、ストレッサーに暴露されても健康的な精神状態を維持する力(抵抗力)と、発病後に健康な状態へと導く力(回復力)、の二側面を持つ概念と捉えられている。しかしながら、従来の精神疾患の神経生物学的基盤研究は、もっぱら脆弱性や発病後の病態を想定したものであり、精神疾患の発症・病態・回復の機序をより深く理解するためには、レジリエンスの側面も取り入れた、一面的でない基盤研究が望まれる。我々は、ストレス脆弱性とレジリエンスの生物学的基盤を解明することでうつ病などのストレス関連精神疾患の発症・回復機序の理解をめざしている。

Stressful events are potent adverse environmental factors that can predispose individuals to psychiatric disorders such as major depression. Exposure to chronic stress can be differently perceived by individuals and can have persistent sequelae depending on the level of stress resilience or vulnerability of each person. While stress vulnerability may influence depression, the molecular and neural mechanisms underlying the susceptibility and resilience to chronic stress within the brain are poorly understood. I focus on the role of epigenetic mechanisms in stress resilience and susceptibility in rodents.
