インフルエンザウイルスのゲノム転写・複製機構の解明
Overview of the research

インフルエンザウイルスのゲノムRNAは、核タンパク質NPやポリメラーゼとともにvRNPを形成する。vRNPはmRNA合成(転写)およびcRNA合成(複製の第一段階)を担う。ウイルス粒子から精製したvRNPは、柔軟な二重らせん構造を持つことが近年の電子顕微鏡解析より明らかとなっている。しかし、mRNAあるいはcRNA合成中のvRNPの構造については全く解明されておらず、また、これらのRNAがどのようにして合成されるのかも分かっていない。そこで私は、高速原子間力顕微鏡(AFM)を用いて機能的な状態のvRNPを解析することによって、微細構造学的観点からインフルエンザウイルスのゲノム転写・複製機構を明らかにすることを目的として研究を行っている。
