The mechanism by which viral sensors activate antiviral interferon responses
研究概要
(日本語)
ヒトを含めた哺乳動物はウイルスの侵入に対して様々な防御機構を持つ。その主となるものが、インターフェロン応答と呼ばれる抗ウイルス自然免疫応答である。自然免疫受容体が、侵入したウイルス核酸を認識することでI型インターフェロン(IFN-およびIFN-)の発現を誘導し、産生されたIFNはその受容体を介してRNaseなどを含む抗ウイルス遺伝子群の発現を誘導し、ウイルス複製を阻害する。ウイルス核酸を認識しインターフェロン産生誘導を担う受容体として主に3つのシステムが知られている。細胞外、特にエンドソームで核酸認識に関わるToll様受容体(Toll like receptors: TLR)、細胞内でウイルスRNAを認識するRIG-I様受容体(RIG-I like receptors :RLR)、そして最近明らかになりつつある細胞内でウイルスDNAを認識するDNA受容体である。これら自然免疫担当受容体、特にRLRを中心としたウイルス認識機構およびインターフェロン応答活性化機構の解明を目的とし日々研究している。
