1. Orderly differentiation system from human pluripotent stem cells (PSCs) towards mesodermal cells and various lineages of blood cells, and induction of PSC-derived hematopoietic stem cells Discovered PET-based collagen sponges allow longer culture times of PSC-derived hematopoietic stem/progenitor cells, leading to increase in the number of them. Established a novel serum-free monolayer culture that can trace the in vivo hematopoietic pathway from ES/iPS cells to functional definitive blood cells via mesodermal progenitors. Further improved, it has been used in all projects in our laboratory to elucidate the pathologies of various diseases through PSC-based disease model. 2. Identification of Functional characters and importance of an extracellular matrix Laminin 411 during vascular endothelial differentiation, and increase in efficacy and purity of iPSC-derived endothelial differentiation. Discovered LM411, a minor extracellular matrix located around blood vessels, is effective at specifying the lineage towards endothelial cells from mesoderm. Furthermore, single-cell RNA-seq study revealed that Rho signals bridge between downstream of LM411 and a common endothelial growth hormone VEGF at the specific stage of differentiation to endothelial cells. 3. Elucidation of the pathology and discovery of novel therapeutic methods of intractable hematological/immunological diseases through PSC-based disease modeling. As a part of AMED Research Center Network for Realization of Regenerative Medicine, drove the studies to explore the pathophysiology of intractable hematological/immunological diseases including Blau syndrome, CINCA syndrome, Fanconi anemia and Severe congenital neutropenia. Contribution: designing experiments, analyzing and preparing data and writing texts. 4. Exploration of the pathology and discovery of novel therapy of hematological malignancy by combining in vivo and in vitro disease models In collaboration with division of Pediatrics, Kyoto University, established the in vivo model of leukemia with poor prognosis to elucidate the pathophysiology especially in extramedullary sites such as liver and central nervous system, leading to discover the niche-like histology and identify the mechanisms in adaptation of leukemic cells in those organs.
