ヒト免疫不全ウイルス(HIV)やヒトT細胞白血病ウイルス(HTLV)など難治性ウイルスの研究を行っている。これらのウイルスは幾つかの共通点がある。すなわち、①長い年月に及ぶ持続感染の末に発症に至ること、②宿主の免疫機構を巧妙に回避出来る技を持っていること、③非常に狭い(選択的な)宿主特異性を示すことである。特にこの宿主特異性(ヒトを含む類人猿にしか感染できない)が、小型実験動物を用いた上記難治性ウイルスのモデル動物研究を困難なものにしている。これまでにウイルスの改変や最適化、宿主の遺伝的背景による選別などにより、実験用霊長類を用いた種々の新規モデルシステムを樹立した。これらを応用して、ウイルスがヒトの体内で引き起こす様々な病態や持続感染のメカニズム、及びそれに対する宿主免疫応答を明らかにするとともに、新規治療法やワクチンの開発といった応用研究も積極的にチャレンジしていきたいと考えている。

Hirofumi Akari is a professor, Center for Human Evolution Modeling Research, Primate Research Institute, Kyoto University since 2009, and also is a concurrent professor, Laboratory of Infectious Disease Model, Institute for Frontier Life and Medical Science, Kyoto University since 2014. He has focused on the cellular and molecular biology of accessory genes of HIV related with viral pathogenesis since he joined the Department of Virology, The University of Tokushima School of Medicine, Japan (Dr. Akio Adachi’s Lab) in 1997 as a research associate and also Viral Biochemistry Section, Laboratory of Molecular Microbiology, NIAID, NIH, USA (Dr. Klaus Strebel’s Lab) in 1999 as a research fellow. He has also been investigating non-human primate models on retroviral as well as flaviviral infection since 2002, which was initiated as the Chief of Laboratory of Disease Control at the Tsukuba Primate Research Center, National Institute of Infectious Diseases (presently, National Institute of Biomedical Innovation), Japan.

ウイルス感染ではその宿主との壮絶な戦いの末に、ウイルスの排除、宿主の死、もしくは共生のいずれかの道へと辿ります。その過程では、例えば宿主は免疫機構によるウイルスの制御、それに対してウイルスは宿主免疫からの回避機構を獲得しその宿主で増殖、そこで宿主はさらに新たな免疫機構の構築、といった終わりなき攻防が繰り返されてきたのです。こうした中で、ある種のウイルスは不幸にしてその宿主の生命をも脅かすような病原性を持つ場合もあります。他方、こうしたウイルスと宿主のせめぎ合いの結果、相互に妥協点を見い出して共存共栄の道に至ることもあるでしょう。 当研究室では、難治性ウイルスであるヒト免疫不全ウイルス、肝炎ウイルス、およびT細胞白血病ウイルスの研究を行っています。これらのウイルスは幾つかの共通点があります。すなわち、①長い年月に及ぶ持続感染の末に発症に至ること、②宿主の免疫機構を巧妙に回避出来る技を持っていること、③非常に狭い(選択的な)宿主特異性を示すこと、などです。特にこの宿主特異性(ヒトを含む類人猿にしか感染できない)のため、小型実験動物を用いたモデル研究が出来ないのです。私達はこの難題に取り組み、これまでにウイルスの改変や宿主の遺伝的背景による選別などにより、実験用霊長類を用いた新規モデルシステムを樹立してきました。 興味深いことに、これらのウイルスは霊長類の進化に合わせて共進化した結果、ヒトに特化した「融通の利かない超精密機器」であることが判ってきました。こうした独特な共進化の仕組みをより詳細に探りながら動物モデルを研究目的に応じて最適化することで、ウイルスがヒトの体内で引き起こす様々な現象や病態を明らかにし、さらに新規治療法やワクチンの開発にも貢献していきたいと考えています。

This laboratory is newly established in 2013 for active and effective collaboration between Institute for Virus Research and Primate Research Institute of Kyoto University. We are interested in the co-evolution between hosts and intractable viruses. Viral infection to a new host may lead to small but life-or-death war, while in due course they could finally compromise and live together (symbiosis). Probably, each battle between viruses and hosts should include a variety of long and exciting story, for example, hosts seek to control viral invasion by innate and acquired immunity and then viruses try to develop new weapon to evade the immunity. Unfortunately, this endless chicken race may accidentally cause viral acquisition of life-threatening pathogenicity to the hosts. Exploring cellular and molecular machinery for the co-evolution will provide us precious hints to survive, either win against viruses or live together. With this in mind, we are investigating the mechanisms for the viral persistency and pathogenesis of intractable viruses, HIV, HTLV, HBV and HCV, by employing non-human primate models for the viral infection.
