Identification of Ras signaling regulators using cDNA expression cloning method
研究概要

マウス線維芽細胞NIH3T3由来でv-Ki-ras遺伝子でトランスフォームしたDT細胞に、cDNA発現ライブラリーをトランスフェクトし、悪性形質が抑制された細胞(リバータント)を得、その中からトランスフェクトしたプラスミドを回収するという発現クローニング法を用いて、がん遺伝子Rasのシグナルを制御する因子の同定を進めている。

To identify new Ras signaling regulators, we have developed a new phagemid cDNA expression vector and constructed cDNA expression libraries for screening flat revertant-inducing genes in v-Ki-ras transformed NIH3T3 (DT) cells. After transfection and the screening, more than 700 flat revertants have been obtained. Currently relatively prominent eighty genes have been identified.
