患者腫瘍由来の癌オルガノイドは、もとの腫瘍の性質を高度に維持した3次元培養システムであり、従来の2次元培養細胞株に代わる疾患モデルとして期待されている。我々が開発したCTOS(Cancer Tissue-Originated Spheroid)法によるオルガノイド培養により、新たな癌の性質の解明や治療ターゲットの開発を目指す。 さらに、癌オルガノイドは症例間の多様性を反映している。この特性を利用し、多数の症例から得られた癌オルガノイドパネルを作成しそれぞれの薬剤感受性を検討することで、治療感受性の予測や薬剤開発スクリーニングへの応用も行っている。

Patient derived cancer organoid is a 3D cell culture system in which cancer cells retain the characteristics of original tumors, thus is a new disease model expected to overcome traditional 2D established cancer cell lines. Using our CTOS (cancer tissue-originated spheroid) method, we are working on elucidating novel nature of cancer cells, as well as developing new target for therapeutic treatment. In addition, patient derived cancer organoids reflect inter-patient heterogeneity of original tumors. Taking advantage of this, we also developing a system to evaluate drug sensitivity which is applicable both for clinical selection of the potentially effective drug for each patient and chemical compound screening during drug development.
