Overview of the research

近年、白内障、アルツハイマー病、動脈硬化症など加齢性の疾患組織内において、異常蛋白質の凝集部位に蛋白質結合型のD-アミノ酸(主としてD-Asp)が多量に発見されている。これら蛋白質中におけるD-Aspの蓄積は加齢による非酵素的な異性化反応の結果である。L-アミノ酸の重合体である蛋白質中にD-アミノ酸が生じ、正常な蛋白質の高次構造が乱されることで不溶化や凝集が生じると考えられる。しかしながら、実際に、タンパク質内にD-Aspが形成されることによる当該タンパク質の機能変化、構造変化を明らかにした研究成果は非常に少ない。このような背景を踏まえ、本研究では、タンパク質内におけるAsp異性化と、それが及ぼすタンパク質の機能低下、構造変化および最終的に引き起こされる加齢性疾患との関連性を明らかにすることを目的としている。

Proteins consist exclusively of L-amino acids; nonetheless, D-aspartic acid (D-Asp) has been detected in various human tissue from elderly individuals. We have developed original D/L analysis systems and identified many isomerization sites in proteins, while pathogenesis has been still obscure. The formation and contribution of D-Asp residues in proteins would be one of significant factors of age-related disease. The goal of our study is to observe actual physiological effect of D-Asp formation in protein, and relate D-Asp residues with age-related diseases.
