生体膜は脂質二重層からなり、その内葉と外葉の間にはリン脂質組成の非対称性を有します。脂質非対称の動的恒常性は、リン脂質を細胞質側に移動するフリッパーゼ(P4-ATPase)とその反対に移動するフロッパーゼ(ABC輸送体)と両方向にかき混ぜるスクランブラーゼによって保持されています。脂質二重層間の脂質組成の時空間的調節は、血液凝固、アポトーシス(細胞死の一種)を起こした細胞の除去、筋細胞の融合、細胞分裂、細胞運動、精子の受精能獲得、細胞内メンブレントラフィックなどに関わることが示唆されていますが、詳細な分子機構はほとんど分かっていません。私たちはP4-ATPaseを中心に、生体膜の脂質二重層間の脂質の量的・質的変化や脂質の時空間的変化が制御する細胞機能の分子機構解明を目指します.

Lipid bilayers, including those of the plasma membrane and the membrane-enclosed compartments along the secretory and endocytic pathways, exhibit asymmetric lipid distributions. The asymmetric distributions are generated and regulated by flippases (P4-ATPases), floppases (ABC transporters), and scramblases. Spatio-temporal regulation of the lipid asymmetry is critical for many biological processes, such as apoptotic cell death, platelet coagulation, fusion of muscle cells, cytokinesis, cell migration, sperm capacitation and intracellular membrane trafficking. However, the molecular mechanism is unknown. We aim to elucidate that how spatio-temporal lipid composition changes are regulated and how such changes regulate cellular function and further homeostasis of organisms.
