X線結晶解析による立体構造解析と生化学的解析により、制限酵素のユニークな特異的塩基配列認識機構および触媒機構を明らかにするとともに、立体構造情報をもとに分子工学的手法を用いてDNA認識特異性が変化した人工エンドヌクレアーゼの構築に成功し、新規なDNAマニピュレーション技術を開発した。
