ミトコンドリアは細胞のエネルギー代謝を支える「発電所」であり、ATP(エネルギー)を産出するための「呼吸鎖酵素」と呼ばれる一連の膜タンパク質群が含まれています。呼吸鎖酵素には数多くの低分子有機化合物が作用することが知られ、医農薬として用いられている「阻害剤」から、基質や抗酸化剤として機能する「ユビキノン」まで、その種類は多岐にわたっています。呼吸鎖酵素群に作用する低分子化合物に注目し、有機化学の視点から個々の呼吸鎖酵素の役割を解明していくことを目指しています。

1. NADH-quinone oxidoresuctase, the first energy-converting enzyme in the respiratory chain, plays a central role in cellular energy metabolism. Using rationally designed analogues of specific inhibitors such as acetogenin, quinazoline, aurachin, and korormicin, we are investigating the mechanism of proton- and sodium- pumping NADH-quinone oxidoreductases (complex I and Na+-NQR, respectively) which remain to be elucidated. 2. Ubiquinone is an essential electron carrier, which functions as a substrate of various redox-enzymes in most prokaryotic and eukaryotic cells. We are synthesizing functionalized ubiquinone probes for identification of novel ubiquinone-binding proteins, as well as characterization of ubiquinone reaction mechanism in the respiratory-enzymes.
