Lineage commitment and pluripotency maintenance in the embryo and stem cells

The aim of my research is to understand the nature of embryo-derived or reprogrammed stem cells to build up the knowledge to ensure the safety of clinical application for stem cell therapy and tissue regeneration. During my research in PhD, I realized that for clinical application of the stem cell therapy, further understanding the mechanisms of cell fate commitment in earlier stage of embryogenesis is a prerequisite, therefore, joined Prof Tam’s lab and commenced my research using epiblast stem cells (EpiSCs) as a tool. EpiSCs are derived from the epiblast of the mouse embryo shortly before the cells lose their pluripotency and undergo cell fate commitment, and are thought to retain some characteristics of the epiblast cells. We performed further characterization of the EpiSCs and identified that i) EpiSCs can be established from wide range of developmental stages but do not maintain the characteristics of their original stages, ii) EpiSCs acquire similar characteristics to late gastrula stage ectoderm as well as anterior primitive streak, and iii) EpiSC lines show different predisposition for germ layer potential and iv) the set of ‘signature’ genes related to germ layer lineages. This study built the benchmark for further understanding the “primed” pluripotency state, different from the “naïve” state of mouse ESCs, but common with human ESCs. The knowledge gleaned from the EpiSCs may be applicable for predicting the lineage potential of hESCs or hiPSCs, and for associating the behavior of these cells in embryo development.
