Namasivayam Ganesh Pandian (Ganesh N. Pandian) completed his schooling at SBOA matriculation higher secondary school and master`s in Biotechnology from D.G.Vaishnav College (University of Madras, India). During the course of his project in Central Leather Research Institute, India, Ganesh joined as project fellow to work with Dr. Anju Chadha, Department of Biotechnology, and in electrical engineering department Indian Institute of Technology – Madras (IIT-M). During his tenure under Dr. Chadha, Ganesh gained experience in multidisciplinary projects involving microbial biotechnology, biosensors and biotransformation. Ganesh was involved in collaborative projects with the TATA chemicals, Malladi drug and pharmaceuticals limited and SPIC pharmaceuticals and Kelvin Chemicals, Chennai. Ganesh got Dainichi foundation scholarship and Monbusho (Japanese government scholarship) for his Ph.D in Graduate school of science and technology, Niigata University and completed his degree in March 2009 with a thesis on `A novel innate immune system in insects` under the guidance of Prof. Hidetaka Hori. Ganesh continued his research as`Special Assistant Professor (Research)` with Prof. Hori. Simultaneously, Ganesh received JST (Japan Science and Technology) fellowship to work as scientific advisor in Ushiki patent office, Japan. Ganesh is now working on developing artificial genetic switches under the esteemed guidance of Prof. Hiroshi Sugiyama as a Program Specific Assistant Professor in Institute for Integrated Cell-Material Sciences (iCeMS), a world premier international (WPI) research center at Kyoto University, JAPAN.

Small Molecules to control cell fate
Overview of the research

Natural epigenetic processes precisely orchestrate the intricate gene network by switching “ON” and “OFF” the genes at the right place and time to regulate cellular homeostasis and phenotype. Epigenetic enzymes like histone deacetylases (HDACs) and histone acetyltransferases (HATs) organize histone acetylation and chromatin architecture to govern endogenous gene expression. Pathological chromatin alterations are inherently flexible and can be reversed through pharmacological intervention. HDAC inhibiting small molecules like suberoylanilide hydroxamic acid (SAHA) have been harnessed to modulate gene expression. However, they operate in a sequence-independent manner (Biotechnol. J., 2012, 7, 798). Taking cues from natural transcription factors, we developed a novel class of compound termed, SAHA-PIP containing sequence-specific pyrrole-imidazole polyamides (PIPs) and SAHA. Screening studies in mouse embryonic fibroblasts (MEFs) showed that certain SAHA-PIPs triggered epigenetic marks associated with transcriptionally permissive chromatin and differentially activated the pluripotency genes like Oct-3/4, Sox2, Klf4 and c-Myc (ChemBioChem., 2011, 12, 2822). Genome-wide gene analysis revealed a SAHA-PIP termed `δ` could multiple pluripotency genes in MEF by more than ten-fold to initiate cellular reprogramming in just 24 h (Sci. Rep., 2012, 2, e544). Evaluation of the effect of SAHA-PIPs on genome-wide gene expression in human dermal fibroblasts (HDFs) divulged that each SAHA-PIP could differentially activate the therapeutically important genes like anti-HIV MX2 and anti-obesity KSR2 (Sci. Rep., 2014, 4, e3843). Among them, the notable `ON` switching SAHA-PIPs in HDFs include `K` for germ cell-associated PIWI pathway genes (Angew. Chem. Int. Ed. 2013, 124, 13652 [HOT PAPER]) and `I` for pluripotency-associated OCT4 pathway genes (ACS Chem. Biol. 2014, 9, 2729). Conjugation of DNA binding domain of `I` with HAT activating N- (4-chloro-3-trifluoromethyl-phenyl)-2-ethoxy-benzamide (CTB) remarkably activated identical cluster of genes as SAHA-PIP `I` to substantiate the role of PIP in sequence-specific gene regulation (Angew. Chem. Int. Ed. 2015, 54, 8700). Since PIP conjugates can be tailored to bind predetermined DNA sequences, strategies to expand DNA-based epigenetic switches could create an epoch-making approach in cellular reprogramming and therapeutic gene regulation.
