I have been researching on Vif (viral infectivity factor), one of the accessory proteins of HIV-1, and the host anti-viral protein APOBEC3, which is counteracted by Vif.

HIV-1のアクセサリー遺伝子の一つであるViral infectivity factor (Vif)はウイルスの自然宿主であるT細胞やマクロファージでの複製に必須である。Vifは宿主のユビキチンリガーゼと複合体を形成し、宿主のウイルス抑制因子であるAPOBEC3ファミリー蛋白群をユビキチン化し、プロテアソーム分解を誘導することでウイルス複製に寄与している。Vifの機能の全貌を分子レベルで解明し、新たな治療法の開発に結び付けることを目標としている。

Viral infectivity factor (Vif), one of the accessary gene of HIV-1, is essential for the viral replication in the natural host T lymphocytes and macrophages. Vif forms a ubiquitin ligase complex with host proteins, and ubiquitinates and induces proteasomal degradation of APOBEC3 family proteins, the host anti-viral factors, leading to the viral replication. I would like to contribute to a novel therapeutics targeting Vif by elucidating all aspects of Vif functions.

VifはHIV-1の複製・伝播に必須のウイルス因子であり、治療標的である。Vifの機能発現に必須の補助因子としてCBF-betaを同定したが、VifとCBF-betaの相互作用も新規の治療標的となりうると考えられる。Vifの機能や細胞内制御の全貌を分子レベルで解明することで、Vifを標的とした新規治療薬の実現に貢献しようと考えている。

Vif is essential for HIV-1 replication and transmission, and a target for anti-HIV-1 therapeutics. I identified CBF-beta as an essential co-factor for Vif function, and the interaction between Vif and CBF-beta should be a novel target for HIV-1 therapeutics. I would like to contribute to a novel therapeutics targeting Vif by elucidating all aspects of Vif functions and intracellular regulations.
